Overview

Dual GIP/GLP-1 receptor agonists activate two incretin hormone receptors at once — glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1). Tirzepatide is the first drug in this class, marketed as Mounjaro for type 2 diabetes and Zepbound for chronic weight management and obstructive sleep apnea.

Simultaneous agonism of the GIP and GLP-1 receptors enhances glucose-dependent insulin secretion, suppresses glucagon, slows gastric emptying, and reduces appetite — producing greater glycemic and weight effects than GLP-1 receptor agonism alone.
